What version of Amplitube are you using? I've not been able to get Amplitube 4 to work. So far as I know there is no 32-bit VST for Amplitube 4. (JK requires 32-bit plug-ins.)
I have been able to get Amplitube 3 to work fine. I put the dll in C:\Program Files (x86)\Vst Plugins. Then scanned for new plug-ins in JK and it picked it up fine. It sometimes takes a bit for JK to load the VSTs when you start a session.
Also, I've read that, for some people, VSTs have to be on the opposite track of the one you want. I've not had that problem. I'm using a Scarlett 2i4 first gen.
