Which is better, a USB connection or coming out of the line out (from your source) with an 1/8" plug for your connection to the interface? Is the USB faster?

Not sure what you'r asking here but a 'pretty standard' signal path would be:
Instrument or microphone via XLR/TS/TRS into your external soundcard > external soundcard via USB (firewire/thunderbolt) into your computer.
